5HT2A polymorphism His452Tyr in a German Caucasian systemic sclerosis population

چکیده
Page 1 of 2 (page number not for citation purposes) Recently, Beretta and colleagues [1] reported a protective association of the serotonin 5-HT2A receptor gene polymorphism His452Tyr (C+1354T, rs6314) with systemic sclerosis (SSc) in an Italian population. 5HT2A accounts for most vasoconstrictive and platelet aggregation due to serotonin activity [2]. Beretta and colleagues also demonstrated that plasma from healthy heterozygous carriers of His452Tyr showed decreased platelet aggregation after serotonin stimulus compared to plasma from healthy individuals homozygous for His452. In consequence, they suggested a functional role of His452Tyr in reducing susceptibility to SSc.

5HT2A polymorphism His452Tyr in a German Caucasian systemic sclerosis population – authors' response
The variance of results between our recent article [1] and the replication study by Kirsten and colleagues [2] in German patients may prompt the reader to ask the simple question: what conclusion can be drawn from these contradictory results? One possible answer is that one of the two studies is wrong and all the clues would indicate the first report as the one describing a spurious finding. Th...
متن کاملA polymorphism in the human serotonin 5-HT2A receptor gene may protect against systemic sclerosis by reducing platelet aggregation
INTRODUCTION Platelet aggregation may contribute to the pathogenesis of systemic sclerosis: following activation, platelets release significant amounts of serotonin - which promotes vasoconstriction and fibrosis, and further enhances aggregation. The C+1354T polymorphism in the exonic region of the serotonin 2A receptor gene determining the His452Tyr substitution was associated with blunted int...
متن کاملMCP-1 −2518 A/G Single Nucleotide Polymorphism in Slovak Patients with Systemic Sclerosis
Recent study in a group of German patients with SSc has implicated the SNP in the MCP-1 gene (-2518 A to G) as a factor of susceptibility to SSc. Reflecting the need for replication of genetic association studies, we investigated if this SNP is associated with SSc in another Caucasian population. MCP-1 -2518 A/G genotypes were determined using PCR-SSP in 46 SSc patients and in 449 healthy subje...

INTRODUCTION Accumulating evidences show that shared autoimmunity is critical for the pathogenesis of many inflammatory rheumatic conditions. Specific phenotype could arise from specific genes, and/or combination of genetic factors and environment. Systemic sclerosis (SSc) belongs to connective tissue disorders and recent data have highlighted strong associations with some autoimmunity genes sh...

ABSTRACT Background: It is well recognized that different patients respond in different ways to medications. The inter-individual variations are greater than the intera- individual variations, a finding consistent with the notion that inheritance is a determinant of drug responses. The recent identification of genetic polymorphisms in drug-metabolizing enzymes and drug transporters led to the ...
